Core science
In vitro plantlets often develop under high humidity, low air movement, limited mechanical load, artificial carbon supply, and low microbial exposure. Leaves and roots formed under those conditions may not immediately regulate water or function like greenhouse-grown tissues.
Acclimatization changes the plant’s atmosphere, substrate, nutrition, irradiance, and microbial environment. Root damage during washing or transfer, poorly aerated media, excessive light, and abrupt humidity reduction can cause losses.
In one drug-type cannabis study, survival differed among hydroponic, peat-plug, and rockwool systems. A newer photoautotrophic study reported direct transplanting without a separate acclimation period for two cultivars under its specific protocol. These findings demonstrate protocol dependence rather than a universal best substrate.
Why this matters in cultivation
- Define transplant readiness, handle roots gently, use a clean but functional substrate, and change humidity and light progressively. Track survival beyond the first upright appearance.

Evidence limits
Acclimation success is genotype-, protocol-, and facility-specific. Survival percentages cannot be transferred without matching the method.
