Core science
In physiological studies, germination commonly ends at radicle protrusion. This macroscopic endpoint can be scored consistently and timed. Production success continues beyond it: the radicle must grow, the hypocotyl must emerge, cotyledons must open, and a normal seedling must establish.
The scoring rule must be written before the test. Define the minimum visible radicle criterion, observation interval, final test time, handling method, and categories for contamination, damage, and abnormal development. More frequent checks improve time resolution. Whether inspection alters moisture or temperature depends on the container and method; closed imaging can avoid that potential disturbance.
Counts should record new events at each interval rather than only cumulative totals. This supports final germination percentage, median or mean time, spread, and comparison among replicates. A seed transferred immediately after scoring must retain its lot and event-time identity if later seedling quality is evaluated.
Why this matters in cultivation
- Separate three acceptance gates: germinated, emerged, and established as a normal seedling. This reveals whether failure occurs before radicle protrusion, during mechanical emergence, or after exposure to the seedling environment.
- Use photographs with scale and IDs for ambiguous cases. Do not repeatedly touch the radicle; it is a delicate growing organ and a major contamination route.

Common misconceptions
Correction: It marks germination, not complete seedling success.
Correction: Lots with the same final result can germinate on very different schedules.
Correction: Handling can damage the root and contaminate the test.
