Terms to know
- surface disinfection
- A controlled treatment intended to reduce microorganisms on exposed tissue surfaces; it does not establish sterility inside the plant.
- endophyte
- A microorganism living within plant tissue without necessarily producing visible symptoms.
- aseptic transfer
- Movement or manipulation of culture material using practices intended to prevent introduction of unwanted microorganisms.
- explant injury
- Damage to cultured plant tissue caused by cutting, handling, chemical exposure, temperature, or other treatment.
- contamination frequency
- The proportion of cultures showing defined contamination within a stated observation interval.
- donor preparation
- Pre-culture management and documentation of source plants intended to improve explant quality and reduce contamination risk before collection.
Core science
Surface disinfection targets organisms on exposed tissue surfaces. It does not reliably eliminate internal endophytes or systemic pathogens. Cannabis nodal explants can carry internal contaminants that emerge after apparently successful surface treatment.
Explant type, age, source position, donor-room sanitation, preconditioning, chemical identity, concentration, exposure, agitation, rinse quality, and operator technique all influence establishment.
More aggressive treatment can reduce contamination while also damaging meristems, leaves, and axillary buds. The useful protocol balances viable clean establishment against tissue injury.
Why this matters in cultivation
- Improve donor cleanliness before increasing chemical severity. Run small controlled trials, include untreated or current-standard comparisons when appropriate, and identify contaminants rather than assuming every cloudy culture has the same cause.
