Terms to know

in vitro
Cultivation of living material under controlled conditions outside the intact organism, commonly in a vessel on a defined medium.
aseptic
Maintained using practices intended to prevent introduction of unwanted microorganisms into a controlled culture.
explant
A piece of plant tissue removed from a donor and introduced into culture.
meristem
A region of actively dividing plant cells that produces new tissues and organs.
nodal culture
In vitro culture initiated from stem tissue containing a node and usually an axillary bud.
micropropagation
In vitro clonal propagation using small plant explants through establishment, multiplication, rooting or elongation, and acclimatization stages.
callus
A mass of relatively unorganized proliferating plant cells that can form at wounds or in culture; callus is not synonymous with roots or shoots.
organogenesis
Development of organized plant organs such as shoots or roots from cultured tissues or cells.
acclimatization
Controlled physiological transition of in vitro plantlets to ex vitro humidity, light, airflow, substrate, and microbial conditions.

Core science

In vitro culture grows plant material in a controlled sterile vessel. The explant may be a node, shoot tip, meristem, leaf, petiole, embryo, or other tissue. The biological response depends on explant type, genotype, medium, plant growth regulators, light, gas exchange, and contamination.

Micropropagation commonly includes donor preparation, culture establishment, multiplication, rooting or elongation, and acclimatization. Some protocols use direct axillary-shoot proliferation; others pass through callus or regeneration pathways that can increase variation risk.

Meristem-tip culture can support pathogen-reduction programs, but success is organism-, cultivar-, and protocol-dependent. A 2025 cannabis study combining meristem-tip culture with thermotherapy reported HLVd eradication in five of thirteen tested cultivars, so meristem-derived material is not automatically HLVd-free or pathogen-free without organism-specific confirmation testing. Clean-stock claims also require identity control.

  • Document every transfer and passage. Keep donor, explant, vessel, medium, operator, date, contamination, growth response, rooting, acclimation, and test records connected through one identifier.

Claim: Tissue culture means genetic engineering.

Evidence limits

Terminology is transferable; recipes are not. Cannabis remains genotype-responsive and many published protocols cover only a few cultivars. Pathogen-reduction claims require organism-specific confirmation testing.
